NEW INDICATION & DOSAGE
Squamous cell carcinoma of the head and neck (SCCHN); used in combination with cisplatin and 5-fluorouracil before patients undergo chemoradiotherapy and surgery--
Adults: 75 mg/m2 as a 1 hour I.V. infusion, followed by cisplatin 75 mg/m2 I.V. over 1 hour on day 1, followed by fluorouracil, 750 mg/m2/day as a continuous I.V. infusion for 5 days; administer every 3 weeks for four cycles before beginning radiotherapy. Or 75 mg/m2 as a 1 hour I.V. infusion on day 1, followed by cisplatin 100 mg/m2 as a 30 minute to 3 hour I.V. infusion, followed by fluorouracil 1,000 mg/m2/day as a continuous I.V. infusion on days 1 through 4; administer every 3 weeks for three cycles before beginning chemoradiotherapy. Dosage may need to be adjusted based on blood counts and serious adverse effects.
