AVAILABLE FORMS
Tablets: 150, 300 mg
INDICATIONS AND DOSAGES
Combination antiretroviral treatment of adults infected with CCR5-tropic HIV-1 detectable, who have evidence of viral replication and HIV-1 strains resistant to multiple antiretrovirals--
Adults: 150 mg P.O. b.i.d. when given with strong CYP3A inhibitors such as delavirdine, ketoconazole, clarithromycin, and nefazodone; 300 mg P.O. b.i.d. when given with tipranavir and ritonavir, nevirapine, enfuvirtide, and other drugs that are not strong CYP3A inhibitors; 600 mg P.O. b.i.d. when given with CYP3A inducers, such as efavirenz, rifampin, carbamazepine, phenobarbital, and phenytoin and without a strong CYP3A inhibitor.
Pediatric patients younger than age 16 years: Safety and effectiveness haven't been established.
CONTRAINDICATIONS AND CAUTIONS
Contraindicated in patients with known hypersensitivity to any component of the drug, in pediatric patients under age 16 years, and in women who are breast-feeding.
Use cautiously in patients with liver disease or co-infection with hepatitis B, and in patients who are at increased risk for cardiovascular events, hypotension, and pregnancy.
INTERACTIONS
Drug-drug.
CYP3A inducers (efavirenz, nevirapine, rifampin): May decrease serum levels and effectiveness if combined with these drugs. Adjust maraviroc dose accordingly.
CYP3A inhibitors (atazanavir, delavirdine, ketoconazole, lopinavir and ritonavir, ritonavir, saquinavir): May increase serum levels and toxicity when combined with these drugs. Adjust maraviroc dosage accordingly.
Drug-herb.
St. John’s Wort: May cause loss of virologic response. Avoid use together.
ADVERSE REACTIONS
CNS: dizziness, fever, paresthesias, dysesthesias, pain, peripheral neuropathies, sensory abnormalities, depression, sleep disturbances.
GI: abdominal pain, constipation, dyspepsia, stomatitis, appetite disorders, hepatotoxicity.
GU: urinary tract symptoms.
Respiratory: cough, URI, sinusitis, bronchitis, pneumonia, bronchospasm.
Other: musculoskeletal symptoms, pruritus, dermatitis, infections, benign skin neoplasms.
Reactions may be common, uncommon, life-threatening,
or COMMON AND LIFE-THREATENING.
